I couldn't tell from the picture which side the white plug was on.
The plugs should really be switched 180 degrees. With the colored plug in the fill hole. Just so you know which cart is which!

To me the coloured plugs look to be much higher than the transparent plugs, so switching the plugs could cause clearance problems. Many newer canon printers have very little clearance over the cartridges.

Then if you have to remove these colored plugs due to clearance problems, then what is their purpose again?

They seal the vent hole when the cart is not being used in the printer (just the same as the tape over the vent hole on bought/OEM cartridges). They don't really need to be coloured.
